EVALUATION OF ANTIMICROBIAL ACTIVITY OF ANTIMICROBIAL FINISH BY ZONE INHIBITION TEST METHOD

INTRODUCTION:
For the evaluation of silver and non silver based antimicrobial finishes, many methods as AATCC test method 100, AATCC test method 147 and so many other qualitative and quantitative test method are used but in this experiment we discussed Zone Inhibition of Microbes on antimicrobial fabric. In the textile and clothing world, finishing plays a vital role for quality and value. Now-a-days textiles and garments consumers are more conscious than ever about their health and hygiene as well as about their surrounding environment. The consumers also desire greater safety and better comfort by avoiding unpleasant odour. The microbial attack is dangerous to both living and non-living matters. Microbial infestations can cause noticeable smell from inner garments such as socks as well as spread of diseases, staining and degradation of textiles and garments, etc. Now-a-days a number of finishing treatments are applied on textiles and garments depending upon the market values and customers taste. Apart from these, the anti microbial finish is considered very important on garment due to its effectiveness towards the quality of garments as well as the hygienic side of the customers.

RECIPE FOR AGAR & BROTH:


Nutrient Broth Nutrient Agar

28 g/l 25 g/l

PROCEDURE:

Prepared the solution of broth by using nutrient broth as 28g/l. Filled 5 cleaned glass bottles up to 9 ml by using the broth solution. Prepared the agar solution by using nutrient agar 25g/l. Sterilized both solutions (broth and agar) at 121C for 15 min by placing in an autoclave machine and then allowed to cool down to room temperature. After cooling 30-35 ml agar solution was put in to two sterilized petri plates and allowed to solidify at room temperature. After cooling the broth bottles to room temperature, the cultured bacteria were introduced to one of the bottles known as 1st bottle and that bottle was placed in oven at 37OC overnight to allow the bacteria to grow.

Then 1ml of solution was taken from 1st bottle and put in to 2nd bottle containing broth. Then 1 ml of solution was taken from 2nd bottle and put in to 3rd bottle containing broth. Then 1 ml of solution was taken from 3rd bottle and put in to 4th bottle containing broth. Then 1 ml of solution was taken from 4th bottle and put in to 5th bottle containing broth.

In this way in the 5th bottle the bacteria solution concentration was decreased from 10^-1 to 10^-4. Then 0.2ml of solution was taken from 5th bottle (10^-4), put in to agar plates and spread evenly on solidify agar. At the end antimicrobial finish treated fabric samples were cut in to coin/round shapes and put into the agar plates. Then these agar plates were put in to oven at 37C overnight. Observations were noted after one day and also after four days. The pictures of the observed results are also given in the report.

RESULTS:

After one day and also after four days zone of inhibition was checked, which is a quantitative method of just knowing as is there bacteria present or not. Inhibition zone formed means finish is bactericides and if not formed then finish is bacteria static.

CONCLUSION:

With the increase in concentration of RUCO-BAC-AGP (silver based antimicrobial finish) and RUCO-BAC-MED (non-silver based antimicrobial finish) the bacterial killing capability of the wound dressing was noticed to be increasing.

The bacterial killing capability of RUCO-BAC-AGP (silver based antimicrobial finish) is greater as compared to RUCO-BAC-MED (non-silver based antimicrobial finish) in all concentrations applied.

In case of 10g/l and 15g/l RUCO-BAC-MED (non-silver based antimicrobial finish) the observed leaching effect may be due to the improper application of finish to fabric.
