1.2 Fundamental requirements for service work

These service instructions describe the CO2 incubator:


• HERAcell® 150 i / 240 i.

Please note that maintenance of the device must be carried out only by
adequately skilled personnel. Prior to repairs, please read these service
instructions carefully.

Skills of service personnel • The replacement of defined spare parts must be performed only by the
Technical Service of Thermo Fisher Scientific or by authorized service
personnel who have been trained by Thermo Fisher Scientific.
• Electrical work must be carried out only by a qualified electrician.
• Work to gas supply lines and to gas supply system components must
be carried out only by trained gas and air conditioning expert person-
nel.
Process procedures for personnel • Service works must be carried out in accordance with the applicable
national regulations.

Process procedures for personnel • For any operation of this device, the service personnel must prepare
clear and concise written instructions in the language of the operating
and cleaning personnel based on the operating instructions, applicable
safety data sheets, plant hygiene guidelines, and technical regulations,
in particular:
– which decontamination measures are to be applied for the CO2
incubator and accessories,
– which protective measures apply when specific agents are used,
– Iwhich measures are to be taken in the case of an accident.
• Repairs to the device must be carried out only by trained and authori-
zed expert personnel.

1.4 Warranty

Requirements for warranty Thermo Scientific warrants the operational safety and functions of the CO2
incubator only under the condition that:
• the device is operated and serviced exclusively in accordance with its
intended purpose and as described in these operating instructions,
• the device is not modified,
• only original spare parts and accessories that have been approved by
Thermo Fisher Scientific are used,
• inspections and maintenance are performed at the specified intervals.
The warranty is valid from the date of delivery of the device to the operator.

1.6 Overview safety notes

The general safety instructions point out potential hazards during repairs.
To prevent constant repetitions, the following safety instructions are not
mentioned explicitly in the individual sections; instead, the sections con-
tain only references to the general safety instructions.
Prior to repairs, read the safety instructions carefully.
For any repairs, the listed location requirements apply.

1.6.1 Safety instructions for repairs


For any repair works at the device, the particular ambient conditions listed
as location requirements apply (see operating instructions).

Contamination hazard!
The incubator can be used for the permanent or temporary
storage of toxic or infectious substances or materials. There-
fore, the device or device components may be contaminated.
Always observe the hygiene regulations of the operator!
• Infectious or toxic substances or residues thereof must
always be removed completely from the work space.
• Prior to repairs, make sure that the device has been cleaned
and disinfected to exclude infection hazards!
• The operator must provide a declaration of decontamina-
tion.

Electric shock!
Contact with current-carrying components may cause a lethal
electric shock.
Prior to repairs, disconnect the device from the power supply
system:
• Isolate the device electrically.
• Protect the device from accidental reconnection.
• Make sure the device is de-energized.
• Ground and short the device.
• Isolate adjacent components or components to repair that
are under voltage.

Handling electrical components!


The electrical components must be handled only in compliance
with the guidelines of IEC-61340 Electrostatic, Handling ESD
(electrostatic discharge). Observe ESD safety measures during:
• packing and storage,
• unpacking,
• installation

Start-up:
If safety appliances were removed or disabled during repairs,
the device must not be started up before the safety appliances
have been reinstalled and checked for proper operation!
Before the incubator is started up and operated, a test run must
be performed.

Recycling:
All components of the device can be recycled.
If toxic or infectious substances were used in the device, the
components must be decontaminated as required before they
are discarded or shipped. For shipped components, a certificate
of decontamination must be attached.

1.6.2 Safety notes on carbon dioxide (CO 2)


Carbon dioxide Since CO2 is rated as a harmful gas, certain safety instructions must be
observed when the CO2 incubator is started up and when the device is
operated.

Suffocation hazard!
Large amounts of CO2 released into the room atmosphere may
cause suffocation.
If CO2 is released, initiate safety measures immediately!
• Leave the room immediately and do not allow others to
enter the room!
• Inform security service or fire department!

Instruction of the service personnel:


Personnel operating devices with CO2 supply must be
instructed about the particularities in the handling of CO2
before starting their work:
• Correct operation of pressurized gas containers and gas
supply systems,
• obligation to report damages and shortcomings in CO2
supply lines,
• measures to be taken in case of accidents or failures.

1.6.3 Safety notes on oxygen (O 2)


Oxygen O2 is a gas that promotes combustion and may explode in combination
with grease-containing materials.

Oxygen explosion!
O2 may explode in combination with oils, greases, and
lubricants. If highly compressed oxygen comes in contact with
grease- or oil-containing substances, the mixture may explode!
• For cleaning these device components, use only oil- and
grease-free lubricants.
• Keep all connections and components of the oxygen system
free from substances that contain oil, grease, or lubricant!

Fire hazard!
Released oxygen promotes combustion. Do not use open
flames in the vicinity of oxygen-operated systems!
• Do not smoke in the vicinity of oxygen systems.

• Do not expose the components of an oxygen system to


excessive heat.

1.6.4 Safety notes on nitrogen (N 2)


Nitrogen Nitrogen mixes easily with air. High concentrations of nitrogen reduce the
oxygen content in the air.

Suffocation hazard!
Large amounts of N2 released into the room atmosphere may
cause suffocation.
If N2 is released, initiate safety measures immediately!
• Leave the room immediately and do not allow others to
enter the room!
• Inform security service or fire department!

3 Installing the covers

3.1 Installing / Removing the control box


cover

The electrical components of the control box [2] are


located underneath the cover [3].
The cover [3] is attached with two hooks [4] to the
guide slots [5] at the rear panel of the device and
secured with 7 retaining screws [1].

Removing the control box cover:


1. Remove the 7 retaining screws [1].
2. Disengage the hooks of the cover [3] from the
guide slots at the rear panel of the device.

Installing the control box cover:


1. Install the cover to the guide slots at the rear
panel of the device.
2. Engage the two hooks [4] of the cover [3] into the
guide slots [5].
3. Secure the cover using the 7 retaining screws [1].

4 Electronic unit components

4.1 Bottle turning device PCB (HERAcell®


240 i)

The bottle turning device PCB [1] is installed in the


control box at the rear panel of the device.

Removing the bottle turning device PCB :


1. Mark the connecting cables for the assignment of
the bottle turning device levels (starting from top:
d, c, b, a).
If required, mark the connections [2] for other
components such as O2 sensor system or gas
monitoring system.
2. Disconnect all connecting cables from the
sockets.
3. Remove the 8 retaining screws and remove the
PCB [1] from the rear panel of the device.

Installing the bottle turning device PCB :


1. Place the PCB onto the screw bases on the rear
panel of the device and secure it using the 8
retaining screws.
2. Connect the connecting cables (see wiring
diagram).

Assignment of the bottle turning device


levels:

Make sure that the bottle turning device
level connecting cables are assigned
correctly to the corresponding sockets a, b,
c, and d.

Checking the bottle turning device speed:


For a start-up of the device after the bottle turning
device PCB has been replaced , the bottle turning
device speeds should be checked.
1. Connect the device to the power supply system.
2. Perform a power reset by switching the device on.
3. In the submenu BOTTLE TURNING DEVICE (optional)
of the user menu OPTIONS, check the bottle
turning device speed on all levels and adjust it as
required (see operating instructions).

5 Sensor system components

5.1 Installing and removing the O2 sensor

The O2 sensor for device version HERAcell® 240 i [1] is


installed to the work space ceiling, the O2 sensor for
device version HERAcell® 150 i [2] is installed in the
upper section of the right sidepanel.
The O2 sensor PCB [6] is installed in the control box at
the rear panel of the device.

O2 sensor control range:


The valid control range of the O2 sensor
and its PCB [6] is defined on the EPROM
[5]!
Control range I: 1% - 21%
Control range II: 5% - 90%

Removing the O2 sensor:


• Pull the sensor [4] out of its base [3].

Installing the O2 sensor:


• Push the sensor into its base.

Replacing the O2 sensor PCB:


To ensure correct function, the O2 sensor
must always be replaced with its PCB as an
assembly!

Removing the O2 sensor PCB:


1. Mark the connecting cables.
2. Disconnect all connecting cables from the
sockets.
3. Remove the 4 retaining screws and remove the
PCB [6] from the rear panel of the device.

Installing the O2 sensor PCB:


1. Place the PCB onto the screw bases on the rear
panel of the device and secure it using the 4
retaining screws.
2. Connect the connecting cables (see wiring
diagram).

5 Sensor system components

Calibrating the O2 sensor:


Calibration For a start-up of the device after the O2 sensor and its PCB have been
replaced the following work steps must be carried out:
1. First check, if the correct control range of the O2 sensor is adjusted.
Request submenu:
 SERVICE LEVEL / FE11 O2 / CONFIGURATION O2.
2. Check calibration of the O2 sensor (see chap. 7.4‚ pg. 41).

5.2 Blower wheel cover removal /


installation

The wire cage [1] that covers the blower wheel [4] is
attached with two screws [5] to the measuring cell [3].
Each screw has a sealing ring that protects it from
getting lost.

Removing the wire cage:


1. Remove the two retaining screws [5] that secure
the wire cage [1].
2. Remove the wire cage.

Installing the wire cage:


1. Install the retaining rings with sealing rings to the
wire cage guide tabs.
2. Align the wire cage so that the retaining screws
[5] are seated at the threaded holes [2] of the
measuring cell [3].
3. Tighten the two retaining screws [5] hand-tight.
The retaining screws must be flush with the
measuring cell.

After installation, check to see if the blower
wheel can rotate freely.

5.3 Installing / Removing the measuring

The blower wheel [3] is secured to its shaft [2] with a


setscrew [2].

Removing the blower wheel


1. Remove the wire cage [6] that covers the blower
wheel (see chap. 5.2‚ pg. 21).
2. Remove the setscrew [4] using an Allen wrench (2
mm).
3. Slide the blower wheel [3] off its shaft.

Installing the blower wheel:


1. Slide the blower wheel [3] onto its shaft.
2. Secure the blower wheel [3] on its shaft. Install the
setscrew [4] hand-tight.
3. Install the wire cage [6] (see chap. 5.2‚ pg. 21).

Functional test
After installation, check to see of the
blower wheel can rotate freely.

6 Service menu operation

6.1 Operational structure service level

The service level is protected by an access code and is


requested in the user menu by pressing the MENU key [1].

6.1.1 Enable the service level


1. Request the dialog box KEYPAD LOCK.

2. Enter the service code:


 999999 ( 6 x numeral 9)
 The number combination is displayed hidden in
the input box [2].

3. Delete the input numbers:


 Press the DELETE key [3] .

4. Exit the menu:


 Press the BACK key [5] .

5. Confirm the input:


 Press the ENTER key [4].
 After the service menus have been enabled
successfully, the USER CONFIGURATION menu is
extended by the option SERVICE LEVEL.

Clearing Service Level:


If the clearing of the Service Level was not
successful, carry out the procedure once
more.
Locking service menus:
After the settings have been completed, a
power reset is performed in the service
menus so that the service mode is locked
again!

6 Service menu operation

6.1.2 Reset of user-defined code


If the user-defined keypad lock code is no longer
available, the code must be reset to the default value:
0000.

1. In the user configuration SETTINGS request sub-


menu KEYPAD LOCK CODE.

2. Enter the reset code:


 8437661 (is equal to: Thermo1 on the
combined editing numeric keypad)
 The number combination is displayed hidden in
the input box [1].

3. Confirm the input:


 Press the ENTER key [2]
 After the default code has been enabled
successfully, the keypad lock can be encoded
again entering: 0000.

6 Service menu operation

6.2 Extension user menus

As long as the service level is enabled, the status bar


[2] indicates SERVICE MODE.
The scope of operation of the user menus is extended
by following the option and functions:

• The USER CONFIGURATION menu is extended by the


submenu SERVICE LEVEL [2].
• The SETTINGS menu is extended by the submenu
TOUCH CALIBRATION [3] (see chap. 7.6‚ pg. 46).
• The O2 SET VALUE menu is extended by the option
CALIBRATION [4].

• The Event logging menu is extended by the sub-


menu DELETE ERROR TABLE [1].
• The RECORDING INTERVAL menu is extended by the
options RESET OF THE RECORDING INTERVALS:
– RESET AUTO-START [2],
– RESET SERVICE [3],
– RESET CONTRA-CON [4].

Automatic service mode reset:


Unless a user action (contact with the
pressure-sensitive areas and keys) occurs
within 30 min., the system automatically
exits the service menu which is then
blanked out as a submenu.
Access must then be reenabled by entering
the code.

6.3 Device configuration menu

If a check or a change of device configuration is


required, in the submenu FE21 DEVICE CONFIGURATION
of the SERVICE LEVEL all adjustments must be
examined.

Procedure:
1. Request the desired submenu for device
configuration.

2. To browse upward in the selection:


 Press the + key [4].

2. To browse downward in the selection:


 Press the – key [2].

3. The new option / value appears in the display [1].

4. To accept and save the selection:


 Press the ENTER key [3].
 The system returns to the FE21 DEVICE
CONFIGURATION menu.
 Press the SAVE key [5] or [6] (depending on the
dialog box to which the submenu is assigned).
 The system performs an automatic power
reset. The power reset is started with a delay of
about 2 seconds. All actions performed
between saving and power reset will be
ignored.

Renew calibrations:
The saving routine will overwrite the exi-
sting calibration values on the main board.
For a start-up of the device:
• Start device using auto-start routine.
• After this renew all calibrations and cor-
responding adjustments for:
- Temperature calibration,
- CO2 calibration,
- if required, O2 calibration.

must be set to 120 V.

default. Otherwise this feature cannot be
selected as a user option.

7 Calibration and settings

7.1 Temperature calibration

A temperature calibration is required, if the difference


between indicated actual value and measured value
(targeting value) is  ±0,2 °C, especially:
• after the main board has been replaced,
• after the TCD type measuring cell has been
replaced,
• after the IR measuring cell has been replaced.

Temperature calibration is performed in two steps:


• Test field calibration on the service level,
• temperature calibration on the user level.

7.1.1 Preparing the temperature calibration


For a comparison measurement, a calibrated
measuring instrument should be used (see chap. 2.2‚
pg. 15).
To minimize temperature fluctuations during the
measurement, the measuring instrument is positioned
in an isothermal container [3] (e.g. a beaker filled with
glycerin) in the work space.
The center of the work space is the reference location
for the comparison measurement.

Isothermal container:
Do not use a container filled with water
as isothermal container as the evaporation
of the water will result in a too low
temperature.
Excessive work space temperature:
Excessive temperature in the work space
after the calibration can be reduced
by opening the doors for approx. 30
seconds.

Alternatively, a temperature probe can be installed at


the same location. The supply line is routed either
through the measurement aperture [2] in the glass
door or through the pipe duct [1] at the rear of the
device.
The center of the work space is again the reference
location.

7.1.2 Temperature test field calibration


The test field calibration is part of the temperature
calibration.

1. Perform a power reset.

2. In the user level main menu, set the set values:


 Desired temperature set value,
 Set the CO2 set value to 0%,
 if the device is equipped with the (optional) O2
control, set the O2 value to 21%.

3. Request the service level.

4. In the service menu FE1 INCUBATION PARAMETERS,


enter the values of the T factors (temperature
factors).
– In the dialog box TEMPERATURE FACTOR
(DEVICE) [1], set the value to 1.000.
– In the dialog box TEMPERATURE FACTOR
(CUSTOMER) [2], set the value to 1.000.

Factor value input:


The temperature factor must be entered
with three decimal places!

5. Perform auto-start.

6. Enable the service level again.

7. After the auto-start routine has been completed,


determine the new temperature factor of the
device. For this purpose, two different procedures
can be used.
– A: Measurement procedure / Temperature
calibration,
– B: Calculation procedure.

7.1.3 A: Measurement procedure


Performing temperature calibration with measured
value:
1. Press the TEMPERATURE DISPLAY key [1].
 The Temperature menu is displayed.

2. Request the Calibration submenu:


 Press the CALIBRATION key [2].

3. Enter the measured value (target value):


The target value can be increased or reduced
gradually. When the corresponding key - [3] or
+[5] is kept depressed, the function switches over
to fast forward; after approx. 3 seconds, the fast
forward speed increases again.

4. Accept and save the target value:


 Press the ENTER key [4].
 The system returns to the main menu.

Setting the new temperature factor of the device:


1. Enable the service level.

2. In the service menu FE1 INCUBATION PARAMETERS,


request the dialog box T FACTOR (CUSTOMER):
 Read and note the value TEMPERATURE FACTOR
(CUSTOMER) [1].

3. Request the dialog box T FACTOR (DEVICE):


 In this box, enter the read and noted value from
TEMPERATURE FACTOR (CUSTOMER) [2].

4. Return to the dialog box T FACTOR (CUSTOMER):


 Set the value TEMPERATURE FACTOR (CUSTOMER)
[3] to 1.000.

7.1.4 B: Calculation procedure


To determine the factor of temperature increase, the
reference value determined in the test measurement is
divided by the actual value measured by the
temperature probe of the device.

1. Read the temperature value:


 Read the reference value at the measuring
instrument,
 read the actual value at the display.

2. Calculate the temperature increase factor:

3. In the service menu FE1 INCUBATION PARAMETERS,


request the dialog box T FACTOR (DEVICE) [1]:
 In this box, enter the factor value.

4. Accept and save the setting:


 Press the ENTER key [2].

7.2 CO2 calibration

A CO2 calibration is required, if the difference between


indicated actual value and measured value (targeting
value) is  ±0,5 %, especially:
• after the main board has been replaced,
• after the TCD type measuring cell has been
replaced,
• after the IR measuring cell has been replaced.

For a comparison measurement (CO2 calibration), a


calibrated measuring instrument should be used (see
chap. 2.2‚ pg. 15).

1. Perform a power reset.

2. In the user level main menu, set the set values:


Press the corresponding displays, then:
 Enter the desired temperature set value,
 set the CO2 set value to 0%,
 if the device is equipped with the (optional) O2
control, set the O2 value to 21%.

3. Perform auto-start.

4. After completion of the auto-start routine check if


the CO2 value is within a range of 0% - 0,2 %.
If not, perform auto-start routine once more.

5. Set CO2 set value to 5 %.

6. Measure the CO2 set value:


 With stable CO2 set value, observe a period of
stability of at least one hour.
 Position the measuring instrument probe in the
work space (see operating instructions) and
determine the measured value.

7. Request the Calibration submenu:


 Press the CO2 DISPLAY [1].
 Press the CALIBRATION key [2].

8. Enter the measured value (target value):


The target value can be increased or reduced
gradually. When the corresponding key - [3] or
+[5] is kept depressed, the function switches over
to fast forward; after approx. 3 seconds, the fast
forward speed increases again.

9. Accept and save the target value:


 Press the ENTER key [4].
 The system returns to the main menu. The
actual value currently measured in the work
space is displayed in the CO2 display field.

7.3 O2 configuration

An O2 calibration is required:
• After an O2 sensor has been replaced,
• after change-over of the O2 sensor to another
control range.

Configuring the O2 sensor:


If the O2 sensor for another control range
has been replaced, prior to performing an
O2 calibration, the O2 sensor must be

1. Enable the service level.

2. In the service menu FE11 O2, request the dialog


box CONFIGURATION O2 [1].

3. Set the control range:


 The control range can be changed by pressing
the key - [2] or +[4].

4. Accept and save the setting:


 Press the ENTER key [3].
 The system returns to the service menu FE11
O2.

7.4 O2 calibration

An O2 calibration is required, if the difference between


indicated actual value and measured value (targeting
value) is:
• in control range I: 1% - 21%:  ± 0,5 %,
• in control range II: 5% - 90%:  ± 2,0 %.

When running auto-start, the O2 value is calibrated to


21% by default. When the O2 calibration is performed
thereafter, this value of 21% is overwritten.
Therefore, if the O2 sensor is to be calibrated:

• This procedure must be performed after the auto-


start routine,
• the calibration must be repeated after each auto-
start routine.

Performing an O2 calibration:
For a comparison measurement (O2 calibration), a
calibrated measuring instrument should be used (see
chap. 2.2‚ pg. 15).

1. Perform a power reset.

2. In the user level main menu, set the set values:


Press the corresponding displays, then:
 Enter the desired temperature set value,
 set the CO2 set value to 0%,
 set the O2 set value:
Control range I: 1% - 21%
Set the O2 value to 5%.
Control range II: 5% - 90%
Set the O2 value to 70%.

3. Position the measuring instrument probe in the


work space.

4. Perform auto-start.

5. Measure the O2 set value:


 After completion of the auto-start routine, with
stable O2 set value, observe a period of stability
of at least one hour.

6. Enable the service level and then request the


CALIBRATION submenu:
 Press the O2 DISPLAY [1].
 Press the CALIBRATION key [2].

7. Enter the measured value (target value):


The target value can be increased or reduced
gradually, pressing the corresponding - key [3] or
+ key [5].
The calibration of O2 control is only possible
within a limited range of ±3 %!

8. Accept and save the target value:


 Press the ENTER key [4].
 The system returns to the main menu. The
actual value currently measured in the work

Determining the cross compensation correction


value:
Cross compensation must be performed only if, with
stable O2 value, the CO2 value is outside a range of
0 %– 0.2% (CO2 value is 0 % and no CO2 is present in
the work space).
Before cross compensation can be performed the
values of the following control systems must be in
well-balanced, stable condition:
• Temperature,
• humidity,
• O2 actual value.

1. In the service menu FE11 O2, request the dialog


box CORRECTION VALUE O2 / N2:
 Press the CORRECTION VALUE O2 key [1].
 The procedure will be performed and the
determined correction values will be displayed
in a dialog box [2].

2. Exit the display:


 Press the END key [3].
 The system returns to the service menu FE11
O2.

7.5 Device configuration settings

Setting the device configuration (see chap. 6.3‚ pg. 28)


is required:
• After the main board has been replaced.

Read the configuration parameters:


If the configuration parameters are still
accessible via the main board, read the
values and write them back onto the main
board after the replacement.

Reading the configuration parameters before the


main board is replaced:
1. Connect The PC to the incubator using RS 232 /
USB interface.
2. Start the PC program HERAcell® 150 i / 240 i and
enable the service windows.
 Request the dialog box GENERAL > PASSWORD:
 Enter the password: elektronik

3. In the dialog box PARAMETER [1], use the option


SAVE TO FILE [4] to save the configuration
parameters into a file.

Restoring the configuration parameters after the


main board has been replaced:
1. Start the PC program HERAcell® 150 i / 240 i and
enable the service windows.
 Request the dialog box GENERAL > PASSWORD:
 Enter the password: elektronik

2. In the dialog box PARAMETER [1], use the option


READ FROM FILE [3] to access configuration
parameters from the file and then activate the
option WRITE ALL PARAMETERS [2] to write the
configuration parameters to the main board.

Check the configuration parameters:


If the main board could not be accessed any longer,
the configuration parameters in the individual
submenus must be checked and if required, must be
set again.

1. In the service menu FE21 DEVICE CONFIGURATION,


request all submenus from both dialog boxes [5]

 Set the values or required options.

2. After finishing the device configuration save the settings:


 Press the SAVE key [6] or [8].
 The system performs an automatic power reset. The power reset is
started with a delay of about 2 seconds. All actions performed
between saving and power reset will be rejected.

Renew calibrations:
The saving routine will overwrite the existing calibration values
on the main board. For a start-up of the device renew all cali-
brations and corresponding adjustments.

Finishing device configuration:


1. Perform auto-start.
2. Perform temperature calibration (see chap. 7.1‚ pg. 35).
3. Perform CO2 calibration (see chap. 7.2‚ pg. 40).
4. If required, perform O2 calibration (see chap. 7.4‚ pg. 41).

7.6 Calibrating TouchScreen

1. Enable the service level.


 The Settings menu is extended by the option
TOUCH CALIBRATION.

2. Calibrate pressure-sensitivity:
 In the SETTINGS menu request the dialog box
TOUCH CALIBRATION [2].
 Follow the instructions at the display and press
the displayed crosses [1] with a blunt pin
running two sensitivity test cycles.

3. After performance of the calibration:


 The determined values [3] are displayed in a
dialog box.
 A reset of the display is performed
automatically.

4. To check the calibration, open a zoom area in the


trend display and check if the touch sensitivity is
suited for capturing and controlling the zoom-in/
zoom-out of the trend diagram (see also
Operating instructions/Handling and control/
Scaling the trend display).
