Coliform is a group of bacteria used as an indicator of pollution, waste and the conditions
were not good for water, food, milk and milk products. This family includes many genera
including Escheriaceae, shigela, Salmonella, Enterobacter, Klebsiella, and Proteus Serrtia as
bacteria causing gastrointestinal infections. This research was conducted to determine the
presence or absence of coliform bacteria in the ice cube home sold processed dikelurahan
meadow expectations Bengkulu city. This research has been carried out in the Laboratory
UPTD city of Bengkulu. Examination using the descriptive analysis portion Coliform MPN
method 5-1-1. Sampling was carried out simultaneously and the method used is total
sampling with a sample size of 16.
The results of this study showed coliform bacteria in ice cube home sold processed
dikelurahan meadow expectations Bengkulu city. The whole ice cubes are sold in village
fields Bengkulu city hopes positive for coliform bacteria (100%).
The results show that the proficiency level of ice in the desert city of Bengkulu expectations
do not meet the standards for the presence of coliform.

RESEARCH METHODS
The place and time of the study
The research was carried on in
UPTD Laboratory Bengkulu City
Health Department in October to May ,
2016.
Population and sample
The population in this study are all
the ice cubes are sold processed home
village of Padang Harapan Bengkulu
city so that the total population of as
many as 16 home selling ice cubes .
Samples in this research is all the ice
cubes are sold in the village meadow
expectations of Bengkulu city ,
bringing the total number of samples
16 .
Examination of the samples
a. Preparation tools and materials
The tools used in this research is
oven ( Olabtech ) , the reaction
tube ( Pyrex ) glass beaker ( Pyrex
) , petridisk , ose or needle
inoculation, Erlenmeyer (Pyrex),
measuring 10 ml pipette (Pyrex),
measuring pipette 1 ml (Pyrex),
Durham tube, sterile bottles
incubator (Nuve), autoclave
(Autoclave), test tube rack, and
light spirits.
Materials and media used to carry
out this research that Lactose
Broth Single Strength (LBSS),
 Lactose Broth Triple Strength
(LBTS), Brilliant Green Broth
lactose (BGLB), aaquadest, and
ice cubes refined home.

number of coliform bacteria can

number of coliform bacteria in

RESULTS AND DISCUSSION
The results of this study showed
16 samples contained coliform
bacteria (100%) which is a strain
Enterohemorhagic (cause of diarrhea
and colitis / bleeding of the colon). It
is seen the gas bubbles produced by
the Durham tube. The presence of gas
bubbles in this test showed that there
 were no coliform bacteria in the ice
cubes.
Discussion
Ice is another form of water in
solid form that occurs when water is
cooled at a temperature of 0C
(273.15K or 3F) at standard
atmospheric pressure. Ice cubes are
considered to extend the shelf life of a
food product, as it relates to the low
temperature of the ice cubes that can
inhibit the growth of microbes. The
freezing process does not destroy
bacteria, many bacteria can survive at
low temperature for a relatively long
period of time and has been found to
be the cause of the explosion in the
digestive tract disease (Sopacua,
2013).
Coliform bacteria is an important
microbiological parameters to see the
quality of the water. Coliform bacteria
group consisting of Escherichia coli,
Enterobacter aerogenes, Citrobacter
fruendi and other bacteria such as
Salmonella typhi, Shigella spp,
salmonella paratyphi and Vibrio
cholerae. Although this type of
bacteria does not cause a particular
disease directly, its presence in water
indicates poor sanitation. Possible ice
cube terkontaminanasi coliform
bacteria because the water used is not
cooked in advance, hygiene time of
making ice cubes less attention, using

not clean.
Water from natural sources can be
drunk by humans, but there is a risk
that this water has been contaminated
by coliform bacteria or harmful
substances. The pathogenic bacteria
form a toxin (poison) after a short
latency period that is several hours can
cause vomiting. And also the water
used does not meet the eligibility
water used to make ice cubes.
Food and beverage processing are
not hygienic and sanitary can lead
their ingredients in foods and
beverages that can cause health
problems to consumers. the higher the
level of contamination of coliform
bacteria is getting higher the risk of
the presence of other pathogenic
bacteria that bias live in human and
animal feces. Coliform bacteria
contained in the water that made for
ice cubes into the causes of diseases
 such as diarrhea, fever, stomach
cramps and vomiting (Zuhri, 2009).
Given the number of ice cubes
used by the public as well as high
interest to consume the ice cubes, the
researchers are interested in doing
research on Coliform Bacteria Test on
Ice Cubes Preparations The housing
sold in the village of Padang Harapan
Bengkulu City in 2016.
CONCLUSION
Based on the findings of coliform
bacteria test on bulk processed Ice
cubes are sold in the Village of Hope
Bengkulu City Padang can be
concluded that:
1. Ice cubes processed village
home sold in Bengkulu City
Padang Harapan entirely
contaminated coliform bacteria.
2. Ice cubes are sold dikelurahan
Bengkulu City Padang Hope
has the highest index value of
265 MPN / 100mL of water,
while the lowest index value
that is 4 MPN / 100mL of
water.
3. From the entire sample ice cubes
are sold in the village of Padang
Harapan Bengkulu City unfit
for consumption.
