EFFECT OF

TEMPERATURE ON
ENZYME ACTIVITY

INTRODUCTION

Enzymes are biological catalysts, or chemicals that speed up the rate of reaction between
substances without themselves being consumed in the reaction.
Enzymes important because it control the speed of chemical reactions in the body.
Without enzyme, these speed of the reactions will take to slow to keep your people alive.
Enzymes are affected by many factors such as temperature, pH, enzyme concentration,
substrate concentration, and the presence of any inhibitors or activators.
The reason to do this lab report is to understand the relationship of the temperature and
the enzyme activity. We can know that how temperature (hot, cold, normal) affect the
activity of enzyme after doing this experiment. On different range of temperature, the
enzyme activity happen differently for example when it is cold temperature, enzymes
happens slowly and take a lot of time to bubbling.
Hydrogen peroxide come from the reaction of oxygen with isopropyl alcohol and
anthraquinone. Hydrogen is not good for people body because it contain a huge amount
of oxygen which the blood cant handle it. Therefore it leads to many potential issues.
Hydrogen peroxide can be broken down by manganese dioxide because it has catalytic
properties and it broke down into water and oxygen. Equation: 2 H2O (aq) (catalase)>
2 H2O (l) + O2 (g)
Hypothesis: The enzyme activity is increased and decreased depend on the hot or the cold
temperature.

MATERIALS
Raw liver
Petri dish
Dropper Pipette
Hydrogen Peroxide solution
Forceps
Liver puree
Glass marking pencil
Ice bath
3 thermometers
25ml graduated cylinder
5 50ml beakers
Filter paper disks
Warm water bath
Time watch
Paper towel

PROCEDURE
Step 1: Use forceps to place a small piece of raw liver in open petri dish. Use a dropper pipette to
put a drop of hydrogen peroxide solution on the liver.
Step 2: Select the proper equipment and technology to measure catalase activity ( filter paper
disk)
Step 3: Use a graduated cylinder to place 25 ml of hydrogen peroxide solution in 50 ml beaker.

Step 4: Use forceps to dip a filter paper disk in liver puree. Wipe out the excess liver on the filter
paper disk by a tissue.
Step 5: Use the forceps to place the filter paper disk on the bottom of the beaker of hydrogen
peroxide solution. Observe the time ( second) it takes to float the filter paper to the top of the
liquid.

When the experiment happen in the hot temperature, the time takes fastest to bubbling
and the amount of bubbles is a lot
When the experiment happen in the room temperature, the time takes faster to bubbling
however it slower than in hot temperature and the bubbles is also a lot but it fewer than in
high temperature.
When experiment happen in the cold temperature, the time takes slowest to bubbling and
the amount of bubbles is not as much as in the room temperature or in the hot
temperature.

DISCUSSION

The trend of the graph is goes down through the temperature develop which mean when
the temperature is raising up, the less time it takes to appear the bubbles. And opposing
site, the low temperature takes more time to bubbles
The reason for we to graph the data is because it more easy for us to read and summarize
the information (the trend) more easier than read it on the table.
The graph support my hypothesis because it said that the high temperature, the activity
happens faster.
Some errors that we faced are we measure the time not correctly and the measure not
accurate which we record the second slower than it exactly happens.
We need to repeat 3 times because we want to have an accurate results. By doing more
experiments, we will have more correct data number.
We have to take the average of these trials because of average is reflect the exact number
of the medium of three trials collaborated. Therefore the result will exactly correct.

RESEARCH QUESTIONS

Enzymes fit together like a form of key - lock modeling. They make the reaction faster.
When the substates go in the enzyme, the form an enzyme - substrate complex.
A change in pH ( acid, base) affect the function of an enzyme, it called denature enzyme
which mean it destroy the characteristic properties and it function cant be reused
anymore.

REFLECTION

I have learnt many new skill such as manage many jobs in the short period of times, I
know how to measure the temperature and also how to record the data. I also can do the
graph and data table which it not similar to me.
I pretty like when I saw the phenomena happens, however I dont like the smell of the
raw liver, seem it pretty stinky and dirty for me to smell it and tidied it after doing an
experiment.
I think my group work organized, we give each members things to do at the beginning of
the experiment. One doing the measurement of the temperature, one take hydrogen
peroxide, one record all the numbers and one tidied up everything after an experiment .
We need to work harder on the next lab experience because our team work sometimes
dont work well because we not really understand each other yet. We should gave each
other jobs to do at first as well as concentrate on the experiments.
