GENERAL INFORMATION

MICROFLOW has been developed for monitoring of air biocontamination level in critical places
(e.g.: clean rooms, food plants, fermentations plants, etc.). The equipment conforms to the new
European ISO/TC 209 dated 25 July 1999 recommendation.
MICROFLOW allows collection of a wide range of airborne biocontaminant by impacting on agar
standard plates (60 mm + 2 mm and 90 mm + 2 mm diameter, filled with every kind of culture
media).
MICROFLOW can be used successfully in a wide range of applications (air quality control in
pharmaceutical, food, cosmetics plants, in hospitals, clinics and laboratories) for sterile or non
sterile areas monitoring. It is very useful also for air monitoring in building air conditioning (HVAC)
plants.


Working principle

The air to be sampled is drawn through a perforated head, at constant velocity and for a period of
time depending on the condition of the area to be controlled. The inflow air impacts in the surface of
the culture media, chosen on the basis of the microbiological investigation to be carried out, placed
in a 90 mm Petri dish. At the end of the sampling period the Petri dish is removed and placed into
an incubator. At the end of the incubation it will be possible to count the CFU/cm
2
and evaluate the
air biocontamination level of the critical area on the basis of air volume sampled .

OPERATIVE INSTRUCTIONS

Preparing the unit

Before to start the sampling, MICROFLOW must be disinfected, programmed, loaded with a plate
and put on a flat surface or on a suitable photographic tripod (avoid to screw too hard).

WARNING: dont screw too hard the tripod to avoid demage

MICROFLOW allows to operate with 60 + 2 mm or 90 mm + 2 mm AGAR PLATES. It depends
on the head assembly chosen.

Before to operate its necessary to set the right plate diameter of the aluminium plate support. This
setting will not be modified until the change of the type or brand of Petri dishes used for the
sampling.


















Placing the plate

Wear sterile gloves. Remove the sampling head handling with care to avoid any contamination of
its surfaces. Deposit the head on a clean surface.

Disinfect the aluminium plate support with a suitable disinfectant, avoiding liquid spillage inside the
instrument.

Place a sterile filled plate on the first aluminium plate
support. Press gently in outer direction and lay the
plate on the others (see figure).Remove the dish lid,
and deposit it on a clean surface.
Close the sampling unit screwing clockwise the
sterilised aluminium head on the plate support.

Setting a sampling program

Switch on the unit by pressing ON push button.

On the display will appear sequentially the following start information:



























FUNCTION: "Sampling"

Press [Start] key to start a sampling using the last stored parameters.

WARNING: using this procedure the delay start is not considered

Press [Prog] key to enter a new sampling program.

Press [Up] and [Down] key to display the following indications :













When the needed sampling is shown, press [Enter].

Run a Manual sampling

The operator can run a manual sampling with times or volumes different from the preset by the
manual equipment control.

To program the Microflow on a manual sampling, display:








and Press [Enter] key.

It will appear:




Where xxx is the last selected flow rate.


If the flow rate is correct, press [Enter]. If not, select the needed flow rate using the [Up] and
[Down] keys. When the flow rate needed is displayed press [Enter].

It will appear:

xxx is the needed flow rate.


If the programmed data sampling are correct, press [Enter] and it will appear the sequence of the
three initial displays. If it is necessary to change some data, press the [Clear] key and the first
display (Manual sampling) will appear.

Press [Start] key to start the sampling. The two red LED will switch on and, on the display, it will
appear:

Press [Stop] key to end the sampling. The two red LED will switch off and, on the display, it will
appear the following message:








A beep will continue until it will not be pressed [ENTER] to store the sampling data or [CLEAR] to
exit without storing .

Run a programmed sampling

To program the Microflow on a programmed sampling, display:









and Press [Enter] key.


It will appear:




xxxx is the last selected volume.




If the volume is correct, press [Enter]. If not, select the needed volume using the [Up] and [Down]
keys, to increase or decrease the value, and the [Set] key to move the cursor.

WARNING: Programmed mode allows to sample air volume between 1 and 2000
Lt. To sample greater air volumes it is necessary to work in Manual
mode.

When the volume needed is displayed press [Enter].

It will appear:





xxx is the last flow rate selected.



If the flow rate is correct, press [Enter]. If not, select the needed flow rate using the [Up] and
[Down] keys. When the flow rate needed is displayed press [Enter].

Set the needed delay using the [Up] and [Down] keys, to increase or decrease the value, and the
[Set] key to move the cursor. When the delay needed is displayed press [Enter].

WARNING: Set EXACTLY delay time (hour an minutes) for the beginning of the
sampling.
If the delay start is not needed, leave 00:00


After the [Enter] key has been pressed, all the parameters will be displayed (Volume, Flow rate,
Sampling time calculated on the volume and flow rate basis, and the eventual delay start)


Where:
Fr = set flow rate (L/min.)
V = sampling volume (Litres)
Ds = delay start
t = sampling duration (the sampling time calculation
is based on the flow rate and volume set)

If the set data are correct, press [Enter] to save the program.
The three first displays will appear.

If it is necessary to change one or more data, press the [Clear] key and it will be shown the initial
display with the programmed sampling settings. Press the [Enter] key up to the parameter that it is
necessary to change. Modify the parameter and press [Enter] to save the setting. Press [Enter]
up to the summing up screen and save the set parameters pressing the [Enter] key.

The three initial displays will appear and the Microflow is ready for the sampling.
Press [Start] key to start the sampling. The two red LED will switch on and, on the display, it will
appear:


a) The delay start is not active:

The two red LED will brighten and, on the display, will be shown:



and then






At the end of the sampling period the red LED will go out and the following message will
appear:











At the same time a beep will inform the user that the sampling is finished. A beep will
continue until it will not be pressed [ENTER] to store the sampling data or [CLEAR] to exit
without storing .

b) The delay start is activated

The two red LED will lighten and, on the display, will be shown:








At the end of the delay start the LED will brighten and, on the display, will be shown:




and then





At the end of the sampling period the red LED will go out and the following message
will appear.










At the same time a beep will inform the user that the sampling is terminated.The beep will
continue until it will not be pressed [ENTER] to store the sampling data or [CLEAR] to exit
without storing .


Run a sequential sampling

To program the Microflow on a sequential sampling, display:









and Press [Enter] key.

If the volume is correct, press [Enter]. If not, select the needed volume using the [Up] and [Down]
keys, to increase or decrease the value, and the [Set] key to move the cursor.


WARNING: The Seqential mode allows ONLY samplings with air volume between
1 and 2000 litres


When the needed volume is displayed press [Enter] key.

It will appear:

[Down] keys. When the flow rate needed is displayed press [Enter].

NUM SQ = number of sequential samplings

BreakSQ = break (minutes) between two
samplings


Set the needed data using the [Up] and [Down] keys, to increase or decrease the value, and the
[Set] key to move the cursor.


WARNING: It is possible to set from 1 up to 99 cycles
It is possible to set a break between 1 and 120 minutes max.


When the sequence needed is displayed press [Enter].

It will appear:









Set the needed delay start for the first sampling using the [Up] and [Down] keys, to increase or
decrease the value, and the [Set] key to move the cursor. When the delay needed is displayed
press [Enter].

After the [Enter] key has been pressed, all the parameters will be displayed (Flow rate, volume, the
eventual delay start, sampling time calculated on the volume and flow rate basis, number of setting
sequences, time between sequences).

t mm:ss = sampling duration in seconds (the sampling time calculation is based
on the flow rate and volume set)

Bseq sss = break between a sampling and the following (seconds)

If the set data are correct, press [Enter] to save the program.
The three first displays will appear.
If it is necessary to change one or more data, press the [Clear] key and the it will be shown the
initial display with the programmed sampling settings. Press the [Enter] key up to the parameter
that it is necessary to change. Modify the parameter and press [Enter] to save the setting. Press
[Enter] up to the summing up screen and save the set parameters pressing the [Enter] key.

The three initial displays will appear and the Microflow is ready for the sampling.

Press [Start] and the Microflow will start the sampling as shown for the programmed sampling.

Between two sequences sampling:


Sq nn/NN = n sampling to do over the total
sampling to do (es : 02/02 says that we
are waiting for beginning the second
sampling up two)
t = countdown end sampling (h/m/s)


In this moment the LED will brighten.


During the following sampling:



Only the number of sequences changes





In this moment the LED will switch on.

At the end of the sampling period the red LED will go out and then it will appear:











At the same time a beep will inform the user that the sampling is terminated. The beep will continue
until it will not be pressed [ENTER] to store the sampling data or [CLEAR] to exit without storing .



End of sampling

Switch off the Microflow pressing the [Off] key.

Remove the sampling head unscrewing anticlockwise.

Close the plate with the lid, and after the identification, bring it to incubation.

Prepare the unit to the next sampling.

WARNING: In order to save energy when MICROFLOW is waiting for a new
sampling, after about 10 minutes of its inactivity the LCD display
lighting system automatically switch off, it will appear the indication
Stand by, until ENTER key is pressed.

When inside the instrument is fitted the download data system (cod. G1117), after the display

FUNCTION: "Battery level"

In the battery level menu:









Press [Enter] and it will appear while the fun switch on:



xxx % battery level

000 % means that the instrument isnt under
recharge



The battery level is shown by a number (%) on by the wording HIGH, WARNING and LOW .

The word WARNING will appear when the level is around 40%, in this case we suggest to recharge
the battery because the instrument will be able to sample for about 20 minutes, lower is the
percentage and lower is the possible working time .
The word LOW will appear when the level arrives to 5%, in this case you have to recharge the
battery before use the instrument, you can only press the [ON/OFF] key.

AQUARIA RECOMMEND TO CHARGE THE BATTERY WHEN IT
ARRIVES IN WARNING LEVEL.

Press the [Clear] key to exit from the Battery level menu.

Press again the [Clear] key to visualize the first three displays.

FUNCTION: "Setup date and clock"


In the Setup date and clock menu,










Press [Enter] after the visualisation and it will appear:










Set data and time using the [Up] and [Down] keys, to increase or decrease the value, and the
[Set] key to move the cursor.

Press the [Clear] key to exit from the Setup date and clock menu.
Press one time the [Clear] key to stay in the Set menu. Press two times the [Clear] key to visualize
the first three displays.


FUNCTION: "Stored samplings"

In the Stored samplings menu:










Press [Enter] after the visualisation an it will appear:


a) Manual sampling

NOTE: during this step, the Microflow has to be connected to a PC with the proper cable. We
suggest to download only after the connection with the PC.
WARNING: Every time the download is done, the Microflow transfer all the data to the
PC. We suggest to erase the data stored on the Microflow after the
download.

FUNCTION: "Delete memory"

In the Delete memory menu








Press [Enter] after the visualisation and it will appear:








Select the starting point of deletion of samplings memorized
Press the [Up] and [Down] keys to scroll the sampling number.
Use [Set] to move the cursor to select the end point of deletion of samplings memorized
Press the [Up] and [Down] keys to scroll the sampling number.

Press [Enter] after the visualisation and it will appear:








If it is not necessary to delete the stored data press the [Enter] key to exit from this menu.
If it is necessary to delete the data press the [Down] key and it will appear:








Press the [Enter] key to delete all the stored data an it will appear








After waiting the time for the memory reset, it will appear automatically the set menu. Press the
[Clear] key to visualise the first three displays.

FUNCTION: "Flow rate calibration"

In the Flow rate calibration menu:









Press [Enter] after the visualisation and It will appear:








In order to avoid an undesired cancellation of the calibration, the password
will be supplied ONLY at the customers who buy the MICROCONTROL II
calibration kit.
Press the [Enter] key to exit from the Flow rate calibration menu.

Press one time the [Enter] key to stay in the Set menu. Press the [Clear] key to visualise the first
three displays.

FUNCTION: " Language Setup "

In the Language Setup menu:









Press [Enter] after the visualisation and It will appear:









Press the [Up] and [Down] keys to scroll the available languages (ITALIANO, ENGLISH,
FRANCAIS, DEUTSCH, ESPANOL).

Press [Enter] to confirm.
After the confirmation, it will appear automatically the set menu. Press the [Clear] key to visualise
the first three displays.

POWER SUPPLY


The unit is powered by a 8 elements Ni/MH battery pack (each element: 1,2V) protect against
short-circuits and termic overcharges.

NOTE: These batteries have no memory effect and can be recharged at any time without
damaging the elements

The chip installed on the instrument avoid to run a sampling if the battery level is not enough to
maintain a constant flow rate as long as necessary


BATTERY CHARGER

a) Sampler switch on but not operative

If the MICROFLOW has been left switched on, although not operating, the battery level will slowly
decrease. If the level decrease over the limit WARNING, will appear:








b) Sampler operative
If MICROFLOW is sampling and during the sampling the battery charge decreases under the
WARNING level (and it is not possible to maintain the constant flow rate), the display will show:

Battery charge

ATTENTION : to avoid any damages to batteries or power adapter we suggest to follow the
following sequence:

- Connect recharger jack to Microflow
- Connect the charger the power supply

After the recharge, disconnect the charger to the power supply then disconnect the jack from the
instrument.

The complete recharging time is about 4-5 hours but it depends on the residual battery charge.

ATTENTION : we recommend to recharge the batteries when the instrument is off.
You can also recharge the instrument switched on but, in this case the charging time could be
longer and the full battery recharge is not guaranteed.

If the Microflow remains connected to the power for more than 8 hours, the charger automatically
switches off.

- Instrument OFF

The charging of the instrument may be followed by observing the LED on the instrument

LED flash rapid: the instrument is in recharge

LED flash slow : the instrument is fully charged (battery level 100%) and in maintenance phase

LED off : the instrument is fully charged (battery level 100%)


- Instrument ON

The LED will be switched off if the instrument is switched on but not active and will display the word
"Battery charging"

ATTENTION : If necessary the Microflow can also be used connected to the power supply
through the battery charger

During the recharging, you can enter the menu and select "level battery charge", on the display will
appear both the charge percentage and the current recharge percentage.
If the charger is connected to the instrument, the battery charge level showed on the display will be
100% (this means that the battery is charging at 100%) while the current recharge will be 100% at
the beginning but it will decrease during the battery charge.
If the charger is not connected to the power supply the current recharge will be equal to 0% (any
current recharge is issued) and the battery charge real percentage value will be displayed.

ATTENTION : To avoid the total batteries discharge, under the threshold value, that would
cause a very long recharge battery time (also over 10 hours), we recommend to recharge the
batteries at least once a month even if the instrument has not been used and in any case, to
recharge always the instrument after the utilisation .

Replacement batteries
To change the batteries is enough to unscrew the metal panel placed at the bottom of the
instrument.
Be careful to avoid lifting the lid to detach the lead from the IR sensor (if something happens
reconnect the cable paying attention to the color of wires).
The batteries are joined to the battery holder by a velcro strip : remove the batteries, disconnect the
battery connector and replaced the batteries.

Head Control - valve
UNIT DISINFECTION


MICROFLOW must be cleaned and disinfected after each sampling cycle to avoid cross
contamination which could modify the reliability of the sampling.

Switch off the unit.

Clean the surface of MICROFLOW using a sterile cloth wet with a suitable disinfectant. After
drying, the unit is ready for a new sampling cycle.

WARNING: DONT SPRAY disinfectant solution directly on the instrument or the
keyboard: these solutions could enter inside the instrument and they
cause damage as example and oxidation of the circuits.

AQUARIA WILL NOT RECOGNIZE THE WARRANTY CAUSED OF
UNSUITABLE DISINFECTANT SOLUTION


The sampling head and the support plate can be sterilized in autoclave, following the standard
procedure of sterilisation ( i.e. 120 C, 18 minute s).

Between two samplings is necessary to disinfect the sampling head with pencil torch supplied with
the starting kit. In the absence of this, it is possible to use a sterile cloth with disinfectant over this. It
is very important to avoid the liquid penetration inside the instrument.

Let dry at room temperature. After drying the sampling head is ready to be used.

Sampling head fire disinfecting

The MICROFLOW kit is equipped with a pencil to disinfect the head. Disinfect always the head
between two samplings to avoid a cross-contamination.











To switch on the pencil torch rotate clockwise the control-valve. When the gas goes out
from the head, bring the fire to light the pencil.

WARNING: Pass quickly the fire on the interior part of the sampling head.
Do not stop the fire on a specific point of the head, it can be damaged.

To switch off the pencil torch rotate counter-clockwise the control-valve.

To recharge the pencil torch it is possible to use the Butan gas used for cigarette-lighters.

Before to recharge the pencil torch be sure it is switched off. The complete recharge needs about
10 seconds. It is necessary to wait some minutes so that the gas is stable.

For correct recharging it is recommended to keep horizontal the pencil torch and the and the
recharge cartridge

All operations shall be performed by qualified personnel only!

There are no parts replaceable by the user himself. In case of problems please call your dealer for
service.

WARNING: It is recommended to calibrate the instrument at least once a
year

In the unit nothing is switched on Verify:

1. Presence of foreign bodies inside the fan
2. If noise still persists, please call for service

Fan velocity is out of any control 1. If the problem is present for a flow rate only,
calibrate that flow rate with the calibration kit
following related instructions.
2. If the problem is present for all flow rates,
verify each flow rate calibration
3. If the model is 60/90, take off the head and
the support plate. Verify the alignment of the
fan
4. If the problem still persists, please call for
service.

WARRANTY



AQUARIA provides 1 year warranty on the instrument starting on the date of purchase.

Within the warranty period Aquaria will eliminate, free of charge, any defects in the MICROFLOW
microbiological air sampler resulting from failures in material or workmanship either by repairing, or
exchanging parts or exchanging the whole appliance as Aquaria may choose.

Damages caused by improper use as well as negligence to use the instrument, normal
deterioration of internal parts, some defects with negligible effects on the ordinary function of the
instrument are not included in this warranty.

This warranty declines in case of technical intervention repairs which are carried out by non
authorized personnel.

Aquaria is not ever responsible, in front of the purchaser or end user, for damage caused by people
or things (included the same instrument) in relation to the functioning of the instrument, that it is
caused by:

a) any type of work or modification carried out inside the instrument unit, or its accessories,
without Aquaria previous authorization and /or Aquaria technical direct instructions, even in
the case that such works are carried out by Aquaria employees, but without Aquaria proper
authorization.

b) to use the MICROFLOW and/or its accessories for purposes which are different from the
original purposes indicated by Aquaria at the time of planning the design, manufacture and
sell the instrument

c) to clean MICROFLOW and/or its accessories with chemical products different from the
Aquaria indications and /or to use Microflow under different operational conditions as
indicated by Aquaria

To correlate the CFUs (Colony Forming Units) present on the agar plate to the most probable
number (MPN) of micro-organisms per cubic meter of air sampled, the following formula is used:
