Objectives: To investigate the role of catalase enzyme and factors affecting how it works. Theory: Catalase is a common antioxidant enzyme which is produced naturally in almost all living organisms. The reactions in catalysis are important to life, for example, it helps the body to break down hydrogen peroxide (a powerful and harmful oxidizing agent) into oxygen and water, this it prevents the accumulation of carbon dioxide bubbles in the blood. Catalase is a very potent enzyme; one molecule of catalase can decompose millions of hydrogen peroxide molecules into oxygen and water. It also uses hydrogen peroxide to oxidize potentially harmful toxins in the body including formaldehyde, formic acid, alcohol, and phenol. Oxygen is crucial to life; however, when we use oxygen our bodies constantly produce free radicals. Free radicals are chemically unstable molecules or atoms. They also make other molecules or atoms in the body very unstable, thus damaging proteins, cell membranes, and even DNA structure. This is a process which can lead to permanent damage to cells and tissues resulting in infection, mental decline, depressed immunity system, joint disease, and heart disease. Free radicals are also considered to play an important role in the aging process. Catalase is constantly in battle against the effect of free radicals to the body. It transforms harmful superoxide radicals into hydrogen peroxide which later breaks down into water and oxygen. Statement of problem: Hypothesis: Tools and ingredients: Tools: 1. 12 pieces of reaction tube 2. 1 piece tube rack 3. Methylated spirit burner 4. 6 pieces of drops pipette 5. 2 pieces of measuring cup 6. Sticks and lighters 7. 1 place mortar Ingredients: 1. Fresh liver extract 2. Heart extract 3. Tuber extract 4. 5. NaCl 10 % 6. NaOH/KOH 10 %

7. Aquadest Steps/How it works: Observation:

No. 1. 2. 3. 4. 5. 6. Liver extract + Liver extract + HCl + Liver extract + NaOH + Heated liver extract + Heart extract + Tuber Extract + Treatment Gas Bubbles ++++ +++ +++ + + Ember ++++ ++ ++ + -

Description: *) Mark: + ++ +++ ++++ Data analysis:

= if it is none = if it is little = if it is average = if it is much = if it is a lot

1. The first tube with liver extract and

produces the most bubbles because there is catalyze

enzyme that functions to neutralize toxic. In this observation, the ember had be burn heavily because the large amount of oxygen causes the large ember. It is caused the large amount of bubble and sees the ember. 2. The second tube with liver extract and and produces much amount of bubbles,

because catalyze enzyme cannot work well in the acidic condition. In other hand the enzyme will work well in the neutral pH. 3. The third tube with liver extract and and produces small amount of bubbles,

because catalyze enzyme cannot work well in the basic condition. In other hand the enzyme will work well in the neutral pH.

4. The fourth tube is being burn with liver extract and

produces small amount of bubbles,

because catalyze enzyme cannot work well in the high temperature (> 40C).The enzyme will be broken. In other hand the enzyme will work well in 30C-40C temperature. Almost enzyme will be broken over the 40C because the protein will clot (coagulation) 5. The sixth tube with heart extract and produces small amount of bubbles, because there

are less amount of catalyze enzyme within heart than in the liver. 6. The seventh tube with tuber extract and produces small amount of bubbles, because

there are less amount of catalyze enzyme within tuber.

Conclusion: 1. Liver produce catalyze enzyme that has function to neutralize toxic in our body. Catalyze enzyme degrease the Hydrogen Peroxide The reaction is 2. The enzymes work is influenced by some factors: a. Temperature The higher temperature, the more active the enzymes work, but in the higher temperature and lower temperature the enzymes activity will decrease. b. pH Catalyze enzyme will only be optimal at the neutral pH. Enzyme will do denaturasi if in acidic or basic condition c. Enzymes concentration Enzyme concentration is linear with the reaction velocity Answer through questions: Bibliography: http://en.wikipedia.org/wiki/Catalase Access Date July 27th 2011 http://www.ehow.com/about_5521462_role-catalase.html Access Date July 27th 2011 Subardi, Nuryani.
