A chemist's quick guide to NMR acronyms and experiments


A collec on of common NMR acronyms are explained below along with a very brief descrip on of the techniques to which they relate. This is designed to help chemists quickly become familiar with terms they may come across in publica ons but may not recognise.

APT: A ached Proton Test An experiment derived from the JMOD experiment and also used for the mul plicity edi ng of carbon13 spectra. The modica on in APT aims to improve the intensity of the slowly relaxing nonprotonated centres by using a smaller ini al excita on pulse to reduce satura on. See also: JMOD DEPT COLOC: Correla on through LongRange Coupling A 2D experiment for determining longrange (2 or 3bond) correla ons between proton and carbon nuclei and thus piecing together organic structures. This carbon detected experiments has largely been superseded by the more sensi ve proton detected HMBC experiment. See also: HMBC HETCOR COSY: Correla on Spectroscopy The original 2D experiment. Used to iden fy nuclei that share a scalar (J) coupling. The presence of odiagonal peaks (crosspeaks) in the spectrum directly correlates the coupled partners. Most o en used to analyse coupling rela onships between protons, but may be used to correlate any highabundance homonuclear spins eg 31P, 19F and 11B. See also: DQFCOSY TOCSY DEPT: Distor onless Enhancement by Polarisa on Transfer A 1D experiment used for enhancing the sensi vity of carbon observa on and for edi ng of 13C spectra. The sensi vity gain comes from star ng the experiment with proton excita on and subsequently transferring the magne sa on onto carbon (the process known as polarisa on transfer). This gain stems from the larger popula on dierences associated with protons, which are four mes those of carbon. The edi ng feature alters the amplitude and sign of the carbon resonances according to the number of directly a ached protons, allowing the iden ca on of carbon mul plici es. The experiment is typically run using dierent nal proton pulse angles (the numbers below), resul ng in diering signs (+ve or ve) for the various carbon resonances: DEPT45 C 0 DEPT90 0 DEPT135 0
