Effect of Growth Hormone on in vitro induction of adventitious buds and shoots and their survival In Pinus roxburghii (Sargent) Abstract

In Pinus species, adventitious bud induction on cotyledons from mature zygotic embryos is the most common method of getting regeneration through organogenesis. we have successfully induced adventitious buds in Pinus roxburghii (Sargent) cotyledons using WPM & DCR media. Parameters were standardized for optimum concentration of BAP needed for the adventitious bud induction and shoot development. For induction of adventitious bud Full strength WPM & Half strength DCR fortified with 2.5 ppm conc. BAP was best suited. Whereas Elongation of buds (shoot induction) was best observed on Full strength WPM & Half strength DCR fortified with 0.5 ppm conc. BAP. Full strength WPM and Half strength DCR basal media with 0.4%AC were effective for elongation of buds to shoots for P. roxburghii. Specifically Full strength WPM were showed better shoot elongation (8.8 6 average shoots per cotyledon) than Half strength DCR medium (4.8 5 average shoots per cotyledon). Many factors either quantitative like light intensity, photoperiod or qualitative factors as Strength of Sterilizing agent, Media Composition & Concentration of Growth Hormone concentration had significant effect on the responses of embryos to the adventitious bud induction & shoot development.
