5.0 RESULTS

5.1 Representative of Agarose gel

5.1.1 Amplification of the STR loci D16S539, D7S820 and D13S317 (SilverSTR® III

marker)

Result of the agarose gel electrophoresis for DNA amplification of the STR

SilverSTR® III marker loci is shown in figure 1. The SilverSTR® III marker loci were

simultaneously amplified. The three loci were amplified properly and all three bands of each

locus can be seen.

5.3 Case study

From figure 1, two samples were chosen for DNA amplification on 1% agarose gel

for SilverSTR® III multiplex loci. The result shows a successful amplification with three

overlapping loci based on the expected sizes. All samples were then subjected to DNA typing

that was performed using 4 % denaturing polyacrylamide gel electrophoresis. The result of

amplification of the loci is shown in Figure 2. The allele classification was used in comparison

with the allelic ladder provided with the STR marker. Direct comparison between the allelic

ladders and amplified samples of the same locus allows for assignment of alleles. The

genotype of the individuals should be determined according to the multiplex primer pairs.

However, the lanes of ladder show faint bands. The bands for all three loci are fuzzy while

there is a locus that does not show bands. The D7S820 does not show any migration of the

samples and no bands can be seen. Thus, the genotype of the samples cannot be determined.
